Pre-Clinical Acute Toxicology Studies

Biomodels offers a simplified pre-clinical non-GLP assessment of acute toxicology to our clients as a first step in establishing the appropriate dose to be given in efficacy studies. These studies are designed to be an early indication of in vivo toxicity. The results of these studies are not intended to support FDA safety claims, but rather to guide early drug development decisions as to appropriate choice of candidate drug formulations. The data obtained here will provide a foundation to establish an in vivo LD50 (lethal dose – 50% of animals) or MTD (Maximum Tolerated Dose). The aim is to evaluate compounds for toxicity in normal mice. Initially, 2 animals will be given a single dose of the drug substance under investigation either based on in vitro toxicity findings (i.e. a TCLD50 in mg/mL translated into mg/kg for the test species), or on data available for similar compounds. Animals are evaluated for signs of acute toxicity and survival for a period of 24 hours. If no toxicity is seen, the dose is increased and the process is repeated, if the initial dose is toxic, the dose is decreased and the evaluation repeated.
